TT病毒 (TTV)为一种新鉴定的输血后肝炎相关的病毒。用聚合酶链式反应 (PCR)扩增TTVDNA序列核苷酸 1915到 2 185之间的片段 ,并将该片段克隆入 pGEM TEasy载体。重组克隆经酶切鉴定后 ,用全自动测序仪测序 ,发现本组病例中 15株TTV毒株间的同源性在 6 6 .8%到99.3%之间 ,与TTV日本株比较同源性在 6 6 .1%到 97.4 %之间 ,分析结果显示在我国不仅有国外已报告的不同基因亚型毒株存在 ,而且 ,分离到的 3株TTV毒株与日本株G1组和G2组的异源性分别达到 13.3% - 33.2 % ,可能为新的TTV基因亚型。
The TT virus (TTV) is a newly identified post-transfusion hepatitis-related virus. The fragment between nucleotides 1915 and 2185 of the TTVDNA sequence was amplified by polymerase chain reaction (PCR) and cloned into the pGEM TEasy vector. The recombinant clones were identified by restriction enzyme digestion and sequenced by automatic sequencing. The homology between 15 strains of TTV strains in this group was between 66.8% and 99.3% In the range of 6.6% to 97.4%, the results of the analysis showed that not only the different subtypes of different genotypes reported in foreign countries were present in our country, but also three isolates of TTV isolated from Japanese strains G1 and G2 Heterogeneity of 13.3% - 33.2%, respectively, may be the new TTV gene subtypes.
