目的探讨神经生长因子(NGF)促进海马胆碱能神经再生与同源盒基因Lhx8之间的关系。方法SD大鼠72只,在制备切割右侧穹隆海马伞模型和NGF侧脑室注射的基础上,分为对照组、切割组、NGF组、切割联合NGF组;Real-time PCR和Western blotting分别检测各组动物海马中Lhx8蛋白的表达变化;免疫荧光技术检测各组动物海马齿状回颗粒下层中胆碱乙酰转移酶(ChAT)/Lhx8双标细胞数。结果切割组和NGF组中Lhx8蛋白的表达量较对照组增高,切割联合NGF组增高更为明显,且Lhx8基因表达也增高;NGF组和切割联合NGF组中海马齿状回颗粒下层(SGZ)中的ChAT/Lhx8双标细胞数也较对照组和切割组明显增多。结论侧脑室注射NGF促进海马胆碱能神经再生与Lhx8的高表达有关。
Objective To investigate the relationship between nerve growth factor (NGF) -induced hippocampal cholinergic nerve regeneration and homeobox gene Lhx8. Methods Seventy two Sprague-Dawley rats were divided into control group, cleavage group, NGF group and NGF group according to the model of right ventricular septal hippocampus and lateral ventricle injection of NGF. Real-time PCR and Western blotting were used to detect The expression of Lhx8 protein in hippocampus of each group was observed. The numbers of ChAT / Lhx8 double-labeled cells in hippocampal dentate gyrus of each group were detected by immunofluorescence technique. Results Compared with the control group, the expression of Lhx8 protein in the cleaved group and the NGF group was significantly higher than that in the control group. The cleavage combined with NGF group was more obvious and Lhx8 gene expression was also increased. In the NGF group and the NGF group, In ChAT / Lhx8 double-labeled cells than the control group and the cutting group increased significantly. Conclusion Intracerebroventricular injection of NGF promotes hippocampal cholinergic nerve regeneration and the high expression of Lhx8.
