目的探讨乳腺非特殊型浸润性导管癌(IDC-NOS)中miR-7表达与临床病理因素,分子特性和预后的关系。方法收集132例IDC-NOS病例和20例正常乳腺组织病例的石蜡标本和临床资料。免疫组织化学检测ER、PR、Ki67和c-erb B-2蛋白表达;荧光原位杂交检测Her2基因扩增;qRT-PCR检测miR-7表达。结果肿瘤大小、组织学分级、TNM分期和肿瘤细胞增殖是IDC-NOS的独立预后因子(P<0.05,P<0.01);腋窝淋巴结转移率高者和Her2基因扩增者的生存时间显著缩短(P<0.05,P<0.01)。miR-7在IDC-NOS中表达较正常乳腺组织低(P<0.01),与淋巴结转移率呈负相关(r=-0.450);miR-7表达与肿瘤组织学分级、TNM分期和预后呈显著负相关(r=-0.236、-0.215、-0.368),且miR-7高表达较低表达者生存时间显著延长(P<0.01)。结论 miR-7表达与IDC-NOS的生物学行为密切相关,可能成为其潜在的预后指标。
Objective To investigate the relationship between miR-7 expression and clinicopathological factors, molecular characteristics and prognosis in non-specific breast invasive ductal carcinoma (IDC-NOS). Methods Paraffin specimens and clinical data of 132 cases of IDC-NOS and 20 cases of normal breast tissue were collected. Immunohistochemistry was used to detect the expression of ER, PR, Ki67 and c-erb B-2 protein; fluorescence in situ hybridization was used to detect Her2 gene amplification; qRT-PCR was used to detect the expression of miR-7. Results Tumor size, histological grade, TNM stage and tumor cell proliferation were independent prognostic factors of IDC-NOS (P<0.05, P<0.01). The survival time of those with high axillary lymph node metastasis rate and Her2 gene amplification was significantly shorter ( P<0.05, P<0.01). miR-7 expression in IDC-NOS was lower than normal breast tissue (P<0.01) and negatively correlated with lymph node metastasis rate (r=-0.450); miR-7 expression was significantly associated with histological grade, TNM stage, and prognosis. Negative correlation (r=-0.236, -0.215, -0.368), and the survival time of miR-7 high expression was significantly prolonged (P <0.01). Conclusion The expression of miR-7 is closely related to the biological behavior of IDC-NOS and may be a potential prognostic indicator.
