目的 探讨NMDA诱导神经元胆囊收缩素mRNA表达及其机制。方法 利用体外培养的胚胎大鼠大脑皮层神经元和NorthernBlot技术 ,观察NMDA诱导神经元胆囊收缩素mRNA的表达以及c -fos、c - jun反义寡聚核苷酸对该作用的影响。 结果 NMDA刺激神经元 2h后 ,胆囊收缩素mRNA的表达量与对照相比升高 317% (P <0 0 1) ,而预先给予 5 0uMol~ 10 0uMol的c -fos、c - jun反义寡聚核苷酸后该作用被明显抑制 (抑制率分别为 71 5 %、2 16 5 % ,P <0 0 1)。结论 NMDA可通过c -fos、c - jun的介导作用诱导神经元合成胆囊收缩素 ,该结果在小剂量NMDA对谷氨酸兴奋性神经毒性的拮抗作用中具有重要意义
Objective To investigate the expression of cholecystokinin mRNA induced by NMDA and its mechanism. Methods The expression of cholecystokinin mRNA in NMDA - induced neurons and the effects of c - fos and c - jun antisense oligonucleotides on the cultured rat embryonic cerebral cortical neurons and NorthernBlot technique were observed. Results After stimulated by NMDA for 2 hours, the mRNA expression of cholecystokinin was increased by 317% (P <0.01) compared with the control, while the expression of c-fos and c - jun antisense oligodeoxynucleotides This effect was significantly inhibited after poly-nucleotides (inhibition rates 71.5%, 216%, P <0.01). Conclusion NMDA induces neuronal synthesis of cholecystokinin through the mediation of c-fos and c - jun, which is of great significance in the antagonism of glutamate excitotoxicity induced by low dose NMDA
