为深化富硒灵芝的基础研究,指导富硒灵芝的生产与应用,以不同含硒量的富硒灵芝子实体为试样,系统地研究了硒在富硒灵芝中的分布,结果表明:富硒灵芝子实体中81.71%~87.92%的硒是以有机硒形式存在,并主要与子实体中的蛋白结合(蛋白硒占总硒的56.0%~61.2%);水溶性蛋白和碱溶性蛋白是灵芝结合硒的主要蛋白,总结合硒量占蛋白硒的75.2%~79.2%。按照结合硒量的多少,硒在蛋白中的分布依次为:水溶性蛋白>碱溶性蛋白>醇溶性蛋白>盐溶性蛋白。此外,应用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)分离富硒灵芝子实体中的含硒蛋白组分,并采用氢化物原子荧光法(HGAFS)对蛋白或多肽条带按分子质量段切割进行硒含量测定,结果表明:在分辨出的13条蛋白条带中,分子质量在8.709~142.53kDa的蛋白或多肽均结合了不同程度的硒,但硒主要分布在分子质量不超过16kDa的蛋白或多肽中。
In order to deepen the basic research of Se-enriched Ganoderma lucidum and guide the production and application of Se-enriched Ganoderma lucidum, the distribution of selenium in Se-enriched Ganoderma lucidum was studied systematically with Se-enriched Semen Ganoderma lucidum fruiting bodies as samples. The results showed that: Among the selenium in Ganoderma lucidum fruiting body, 81.71% ~ 87.92% selenium exists in the form of organic selenium and mainly binds to protein in fruiting bodies (selenium accounts for 56.0% ~ 61.2% of total selenium); water-soluble and alkali-soluble proteins are Ganoderma lucidum combined with the main selenium protein, the total combined selenium accounted for 75.2% to 79.2% of protein selenium. According to the amount of selenium, the distribution of selenium in protein is: water-soluble protein> alkali-soluble protein> alcohol-soluble protein> salt-soluble protein. In addition, the selenoprotein components of Se-enriched Ganoderma lucidum fruiting bodies were separated by SDS-polyacrylamide gel electrophoresis (SDS-PAGE), and the protein or polypeptide bands were separated by molecular mass section using hydride atomic fluorescence spectrometry The results showed that the proteins or peptides with the molecular mass of 8.709 ~ 142.53kDa all combined with different levels of selenium in the identified 13 protein bands, but the selenium mainly distributed in the molecular mass of not more than 16kDa Protein or polypeptide.
