目的探讨富含Trp/Arg六肽对糖尿病小鼠皮肤创伤愈合的影响。方法经小鼠腹腔注射链脲佐菌素建立小鼠糖尿病模型,在糖尿病小鼠背部切取直径8 mm的圆形全层皮肤,形成皮肤损伤模型。将皮肤损伤模型小鼠随机分为富含Trp/Arg六肽RRWWCR、RRWWFR、RRWWTR组和生理盐水组,分别在创伤形成同时,在伤口处擦拭0.1 mg/ml相应药物,每只0.2 ml,开放式治疗10 d,每天1次。每天观察创面形态变化,并测量创面直径;每5 d取附带周围正常组织的创面全层皮肤,进行组织病理学观察。结果 RRWWCR和RRWWTR可促进创面组织的修复,表现为加强早期炎症反应、促进新生血管、肉芽组织和再上皮化形成以及缩短创面愈合时间。结论富含Trp/Arg六肽对糖尿病小鼠皮肤创伤愈合具有促进作用,其第5位氨基酸残基极性可极大地影响其促修复能力。
Objective To investigate the effect of Trp / Arg hexapeptide on skin wound healing in diabetic mice. Methods Mouse diabetic model was established by intraperitoneal injection of streptozotocin in mice. A circular full-thickness skin with a diameter of 8 mm was excised from the back of diabetic mice to form a skin injury model. The skin lesion model mice were randomly divided into RRWWCR, RRWWFR, RRWWTR group and saline group which were enriched in hexapeptide of Trp / Arg, and wiped the corresponding drug 0.1 mg / ml 0.2 mg Treatment for 10 days, 1 day. The morphological changes of the wounds were observed every day, and the diameter of the wounds was measured. The full-thickness skin of the wounds with the surrounding normal tissues was taken every 5 days for histopathological observation. Results RRWWCR and RRWWTR promoted the repair of wound tissue by enhancing the early inflammatory response, promoting the formation of neovascularization, granulation tissue and re-epithelialization and shortening the wound healing time. Conclusion Enrichment of Trp / Arg hexapeptide can promote skin wound healing in diabetic mice, and the fifth amino acid residue polarity can greatly affect its promoting repair ability.
