将EB病毒感染的人胎儿鼻咽粘膜移植于22只Balb/c裸鼠皮下,每周一次皮下注射丁酸和佛波醇二酯(TPA)。约10天后肿物逐渐增大,15周内行病理学检查,诊断为恶性淋巴瘤4例(其中T淋巴细胞型3例),未分化癌3例。PCR和原位杂交显示,两种肿瘤组织和细胞中均含有EB病毒LMP1基因,核苷酸序列分析表明,与B95一8细胞LMP1基因相应序列的同源性约为96%和99%。说明EB病毒感染人胎儿新鲜鼻咽上皮细胞和淋巴细胞,引起细胞恶性转化后,EB病毒DNA仍存在于癌细胞中。本研究在国际上首次证明,完整的EB病毒和促癌物协同作用可以诱发未分化癌。
The EB virus-infected human fetal nasopharyngeal mucosa was transplanted subcutaneously in 22 Balb / c nude mice and the mice were injected subcutaneously with butyrate and phorbol diester (TPA) once a week. About 10 days after the tumor gradually increased, 15 weeks pathological examination, diagnosis of malignant lymphoma in 4 cases (including 3 cases of T lymphocyte type), undifferentiated carcinoma in 3 cases. PCR and in situ hybridization showed that the Epstein-Barr virus LMP1 gene was contained in both tumor tissues and cells. Nucleotide sequence analysis showed that the homology of LMP1 gene was 96% and 99% with that of B95-8 cells. EB virus infection in human nasopharyngeal epithelial cells and lymphocytes, causing malignant transformation of cells, Epstein-Barr virus DNA is still present in cancer cells. This study for the first time in the world proves that the complete synergism of Epstein-Barr virus and carcinogen can induce undifferentiated carcinoma.
