目的 :探讨缺氧诱导因子 1α(HIF 1α)的表达对结直肠癌血管生成的影响及HIF 1α表达和微血管密度 (mi crovesseldensity ,MVD)与结直肠癌生物学行为的关系 .方法 :采用免疫组织化学方法检测 6 1例结直肠癌组织中HIF 1α表达 ,并用CD34抗体标记血管内皮细胞 ,计数结直肠癌组织中MVD .结果 :MVD的x±s为 (2 8.7± 1 2 .9) (6~5 5 ) ,HIF 1α阳性表达率为 6 5 .6 % ,其中弱、中、强阳性表达率分别为 2 8%、2 0 %、1 8% .HIF 1α表达及MVD与肿瘤浸润深度、淋巴结转移、肝转移和Dukes分期密切相关 (P <0 .0 1 ) .MVD与HIF 1α阳性表达程度成正相关 (rs =0 .795 ,P <0 .0 1 ) .结论 :结直肠癌血管生成与HIF 1α表达密切相关 .HIF 1α过表达及高MVD与结直肠癌的不良生物学行为有关 ,可能成为预测结直肠癌侵袭转移的重要指标
Objective: To investigate the effect of hypoxia inducible factor 1α (HIF-1α) on the angiogenesis of colorectal cancer and the relationship between HIF-1α expression and microvessel density (MVD) and the biological behavior of colorectal cancer.Methods: The expression of HIF-1α in 61 colorectal cancer tissues was detected by chemical method, and the vascular endothelial cells were labeled with CD34 antibody, and the MVD in colorectal cancer tissues was counted.Results: The X ± s of MVD was (2 8.7 ± 1.29) (6 ~ 5 5), the positive expression rate of HIF 1α was 65.6%, of which the expression rates of weak, moderate and strong positive were 28%, 20% and 18% respectively.HIF 1α expression and the depth of MVD and tumor infiltration, Lymph node metastasis, liver metastasis and Dukes stage (P <0.01) .MVD was positively correlated with the positive expression of HIF-1α (rs = 0.795, P0.01) .Conclusion: Colorectal cancer angiogenesis Is closely related to the expression of HIF-1alpha.HIF-1alpha overexpression and high MVD are associated with poor biological behavior of colorectal cancer, which may be an important index to predict the invasion and metastasis of colorectal cancer
