目的:观察山楂叶总黄酮(hawthorn leaf flavonoids,HLF)对心肌缺血大鼠心电图、心室/体质量指数、血清肌酸激酶(CK)、乳酸脱氢酶(LDH)活性及脂质过氧化物丙二醛(MDA)含量的影响,探讨HLF作用机制。方法:将大鼠随机分为4组:正常对照组,急性心肌损伤模型组,HLF高(52mg.kg-1.d-1)、低(26mg.kg-1.d-1)剂量组。HLF组腹腔注射HLF7d,在第4天,与模型组一起连续皮下注射ISO150mg.kg-1.d-13d。正常对照组腹腔/皮下注射等体积的生理盐水。最后一次注射ISO24h后,检测每组大鼠Ⅱ导联心电图、心室/体质量指数、血清CK、LDH活性及MDA含量。结果:ISO模型组死亡率高(56.2%),存活大鼠ST段显著上抬,心室/体质量指数、血清CK、LDH活性及MDA含量均增加。而HLF组死亡率显著降低,ST段、心室/体质量指数、血清CK、LDH活性及MDA含量均趋于正常。结论:HLF能有效防止ISO诱导的心肌损伤,机制可能与其抗氧化作用有关。
Objective: To observe the effects of Hawthorn leaf flavonoids (HLF) on electrocardiogram, ventricular/body mass index, serum creatine kinase (CK), lactate dehydrogenase (LDH) activity and lipid peroxidation in rats with myocardial ischemia Effect of malondialdehyde (MDA) content on the mechanism of action of HLF. Methods: Rats were randomly divided into 4 groups: normal control group, acute myocardial injury model group, HLF high (52mg.kg-1.d-1), low (26mg.kg-1.d-1) dose group. The HLF group was intraperitoneally injected with HLF for 7 days. On the 4th day, the rats were injected subcutaneously with the model group with ISO 150mg.kg-1.d-13d. In the normal control group, an equal volume of physiological saline was intraperitoneally/subcutaneously injected. After the last injection of ISO24h, the II lead electrocardiogram, ventricular/body mass index, serum CK, LDH activity and MDA content were measured in each group. RESULTS: The mortality was high in the ISO model group (56.2%), the ST segment of the surviving rats significantly increased, and the ventricular/body mass index, serum CK, LDH activity and MDA content all increased. On the other hand, mortality in the HLF group was significantly lower, ST segment, ventricular/body mass index, serum CK, LDH activity, and MDA content all tended to be normal. Conclusion: HLF can effectively prevent ISO-induced myocardial injury, and the mechanism may be related to its anti-oxidation effect.
