目的:观察益气化瘀清热方及其拆方各类含药血清对脂多糖(lipopolysaccharide,LPS)诱导下体外培养大鼠肾小球系膜细胞(primary mesangial cells,Ms C)表达转化生长因子βI型受体(transforming growth factorβ,TGF-β-RI)mRNA及其蛋白的影响。方法:采用RT-PCR法检测各组含药血清对Ms C表达TGF-βRⅠ mRNA的影响;采用Wersten Blot法检测各组含药血清对大鼠MsC表达TGF-βRⅠ蛋白的影响。结果:(1)RT-PCR结果显示:各组含药血清均能抑制体外培养的Ms C表达TGF-βRⅠ mRNA(P<0.01),复方组作用最强,与其他含药血清组比较差异显著(P<0.01),清热组与化瘀组比较无明显差异(P>0.05),但优于益气组(P<0.01)。(2)Wersten印迹结果显示:各组含药血清均能抑制体外培养的Ms C表达TGF-βRⅠ蛋白,复方组作用最强,与其他含药血清组比较差异显著(P<0.01),清热组与化瘀组比较无明显差异(P>0.05),但优于益气组(P<0.01)。结论:益气化瘀清热方及其拆方、雷公藤多苷的含药血清均能抑制体外培养的大鼠Ms C表达TGF-βRⅠmRNA及其蛋白。复方组作用较强,清热组及化瘀组次之,益气组最弱。
OBJECTIVE: To observe the effects of Yiqi Huayu Rejie Decoction and its disassembled prescriptions on the expression of transforming growth factor-β (TGF-β) in primary cultured rat mesangial cells (MsCs) induced by lipopolysaccharide (LPS) β I receptor (TGF-β-RI) mRNA and its protein. Methods: RT-PCR was used to detect the effect of each serum group on the expression of TGF-βRⅠmRNA in MsC group. The Wersten Blot method was used to detect the effect of each serum group on MsC expression of TGF-βRI. Results: (1) The results of RT-PCR showed that the drug-containing serum of each group could inhibit the expression of TGF-βRⅠmRNA in Ms C (P <0.01), the effect of compound group was the strongest, the difference was significant compared with other drug-containing serum groups (P <0.01). There was no significant difference between heat-clearing group and blood-stasis group (P> 0.05), but it was better than that of Yiqi group (P <0.01). (2) The Wersten blotting results showed that all the drug-containing serums could inhibit the expression of TGF-|ÂRI protein in MsC cells in vitro, and the effect of compound group was the strongest, with significant difference compared with other drug-containing serum groups (P <0.01) There was no significant difference with Huayu group (P> 0.05), but superior to Yiqi group (P <0.01). CONCLUSION: Yiqi Huayu Qingre prescription and its disassembled prescriptions and the drug-containing serum of tripterygium glycosides can inhibit the expression of TGF-βRⅠmRNA and its protein in Ms C cultured in vitro. The effect of compound group is stronger, followed by heat-clearing group and blood-stasis group, and weakest by Yi-Qi group.
