目的用衍生化法测定山药中还原型谷胱甘肽(GSH)和总巯基(-SH)的含量。方法利用4,4′-二硫代联吡啶(4-DPS)和邻苯二甲醛(OPA)为衍生化试剂,采用细胞破壁和衍生化为一体的样品前处理方法,分别用UV-vis分光光度法和荧光光谱法测定了山药鲜品中总巯基(-SH)、GSH的含量。结果山药中含较丰富的总巯基和GSH,含量分别达0.4556μmol/g和0.1792μmol/g。UV-vis分光光度法和荧光光谱法测定方法的检测限分别为1.0458μmol/L和0.5666μmol/L,回收率分别为98.73%和98.40%。结论本研究对山药中巯基化合物的含量、测定方法学和样品前处理方法提供了重要的基础数据和参考依据。
Objective To determine the content of reduced glutathione (GSH) and total thiol (-SH) in yam by derivatization. Methods 4,4’-dithiodipyridine (4-DPS) and phthalaldehyde (OPA) were used as derivatization reagents. Cell-wall and derivatization were used as sample pretreatment methods. UV-vis The content of total sulfhydryl (-SH) and GSH in yam fresh product was determined by spectrophotometry and fluorescence spectrometry. Results Yam was rich in total sulfhydryl and GSH contents of 0.4556μmol / g and 0.1792μmol / g, respectively. The detection limits of UV-vis spectrophotometry and fluorescence spectrometry were 1.0458μmol / L and 0.5666μmol / L, respectively. The recoveries were 98.73% and 98.40% respectively. Conclusion This study provides important basic data and references for the content of sulfhydryl compounds in yams, determination methods and sample pretreatment methods.
