目的:探讨caspase-6对成骨肉瘤细胞株的凋亡诱导作用。方法:应用RT-PCR方法检测成骨肉瘤细胞株SOSP-9607中caspase-6基因的表达水平。以腺病毒adv5作载体,构建含caspase-6基因的转基因载体,用脂质体包裹法转染SOSP-9607细胞株,用RT-PCR方法检测转染后SOSP-9607细胞中caspase-6基因的表达。用MTF法检测转染caspase-6对SOSP-9607细胞株生长的影响。结果:成骨肉瘤细胞株SOSP-9607中未检出caspase-6表达。RT-PCR法证实转染。caspase-6后SOSP-9607中caspase-6表达显著增强,MTT检测显示对SOSP-9607细胞的生长有抑制作用,形态学观察及DNA电泳证实转染后的细胞株存在细胞凋亡特征。结论:caspase-6对成骨肉瘤细胞的生长有抑制作用,并可诱导成骨肉瘤细胞凋亡。
Objective: To investigate the apoptosis-inducing effect of caspase-6 on osteosarcoma cell lines. Methods: The expression of caspase-6 gene in osteosarcoma cell line SOSP-9607 was detected by RT-PCR. The adenovirus adv5 was used as the vector to construct the transgene vector containing the caspase-6 gene. The SOSP-9607 cell line was transfected by lipofectamine. The expression of caspase-6 gene in SOSP-9607 cells was detected by RT-PCR expression. The effect of transfection of caspase-6 on the growth of SOSP-9607 cell line was detected by MTT assay. Results: The expression of caspase-6 in osteosarcoma cell line SOSP-9607 was not detected. Transfection was confirmed by RT-PCR. The expression of caspase-6 in SOSP-9607 cells was significantly increased after caspase-6 treatment. MTT assay showed that SOSP-9607 cells could inhibit the growth of SOSP-9607 cells. Morphological observation and DNA electrophoresis confirmed the existence of apoptotic cells. Conclusion: Caspase-6 inhibits the growth of osteosarcoma cells and induces osteosarcoma cell apoptosis.
