目的探讨多重引物PCR和异源双链毛细管电泳技术(heteroduplex analysis HA)在B细胞淋巴瘤诊断中的应用。方法从普通石蜡切片组织中提取基因组DNA用多重引物PCR扩增。利用双链异源技术进行毛细管电泳,并对其产物进行分析。结果 32例B细胞淋巴瘤中29例检测有克隆性重排,检出率90.65%。10例反应性增生病例均成多克隆性重排。结论在B细胞受体IgH和IgK基因克隆重排检测中,多重引物PCR及异源双链毛细管电泳技术具有样本来源便利,灵敏度高,特异性强,耗时短等特点,可作为B细胞淋巴瘤的诊断及鉴别诊断的辅助检测手段。
Objective To investigate the application of multiplex primer PCR and heteroduplex analysis HA in the diagnosis of B cell lymphoma. Methods Genomic DNA was extracted from normal paraffin sections and amplified by multiplex PCR. The use of double-stranded heterologous technology for capillary electrophoresis, and its products were analyzed. Results Twenty - nine cases of B - cell lymphoma detected clonal rearrangements with the detection rate of 90.65%. 10 cases of reactive hyperplasia cases were polyclonal rearrangements. Conclusions Multiplex PCR and heteroduplex capillary electrophoresis with convenient source, high sensitivity, high specificity and short time-consuming in multiplex cloning rearrangement of IgH and IgK gene of B cell receptor can be used as B cell lymph Tumor diagnosis and differential diagnosis of the auxiliary detection means.
