评价补阳还五汤及其精简方(脑健片)对脑缺血体内模型大鼠海马组织周期蛋白CDK4/Cyclin D1的调控。将SD大鼠随机分为假手术组、模型组、补阳还五汤干预组(灌胃3.15 g·kg-1)及补阳精简方干预组(灌胃2.41 g·kg-1),每组按给药后1,3,7,14,28 d再各分5组。采用免疫组化法和实时荧光定量PCR法考察不同时间点各组大鼠海马组织中CDK4/Cyclin D1的表达。结果发现脑缺血模型大鼠海马组织中,CDK4/Cyclin D1的表达上调,与假手术组比P<0.01,提示脑缺血损伤将激活细胞周期信号通路;补阳还五汤及其精简方干预后的3,7,14,28 d均能显著降低周期蛋白的表达,与模型组比P<0.05,提示补阳还五汤及其精简方均可通过细胞周期信号通路对神经细胞发挥保护作用。
To evaluate the effect of Buyang Huanwu Decoction and its concise prescription (BSJ) on the expression of CDK4 / Cyclin D1 in hippocampus of cerebral ischemia model rats. The SD rats were randomly divided into sham operation group, model group, Buyang Huanwu Decoction intervention group (intragastric administration of 3.15 g · kg-1) and BuYangJianJian intervention group (intragastric administration of 2.41 g · kg-1) Groups were divided into 5 groups after 1, 3, 7, 14 and 28 d after administration. The expression of CDK4 / Cyclin D1 in hippocampus of rats at different time points was examined by immunohistochemistry and real-time fluorescence quantitative PCR. The results showed that the expression of CDK4 / Cyclin D1 was up-regulated in hippocampus of rats with cerebral ischemia, which was significantly lower than that in sham-operation group (P <0.01), suggesting that cerebral ischemia injury could activate cell cycle signaling pathway. Buyanghuanwu Decoction After 3, 7, 14, and 28 days, the expression of cyclin was significantly decreased compared with the model group (P <0.05), suggesting that Buyang Huanwu Decoction and its abbreviated prescription can all play a protective role on nerve cells through the cell cycle signaling pathway effect.
