目的建立柱前衍生化HPLC法测定鸦胆子油中油酸和亚油酸的含量。方法将鸦胆子以石油醚回流提取得到鸦胆子油,经柱前衍生化后,利用HPLC法测定油酸和亚油酸的含量。色谱条件:色谱柱为WatersSymmetry C1(8150 mm×3.9 mm,5μm),检测波长242 nm,流动相为甲醇-水(98∶2),柱温:28.5℃,流速:1.0 mL.min-1。十七烷酸为内标。结果油酸在0.04~0.8 mg.mL-1,亚油酸在0.02~0.6 mg.mL-1范围内具有良好的线性关系。平均加样回收率油酸为99.04%,亚油酸为96.97%。在6 min内可完成一个样品的分析。结论本法操作简便,分析时间短,为评价和控制鸦胆子油质量提供了依据。
OBJECTIVE To establish a pre-column derivatization HPLC method for the determination of oleic acid and linoleic acid in Brucea javanica oil. Methods Brucea javanica was extracted with petroleum ether and refluxed. Brucea javanica oil was obtained after derivatization with propidium iodide. The contents of oleic acid and linoleic acid were determined by HPLC. Chromatographic conditions: Waters Symmetry C1 (8150 mm × 3.9 mm, 5 μm) was used with a detection wavelength of 242 nm. The mobile phase consisted of methanol-water (98: 2) with a column temperature of 28.5 ℃ and a flow rate of 1.0 mL · min-1. Heptadecanoic acid is an internal standard. The results showed that oleic acid had a good linear relationship in the range of 0.04-0.8 mg.mL-1 and linoleic acid in the range of 0.02-0.6 mg.mL-1. The average recovery of oleic acid was 99.04% and linoleic acid was 96.97%. A sample analysis can be done within 6 minutes. Conclusion This method is simple, short analysis time, which provides the basis for evaluating and controlling the quality of Brucea javanica oil.
