已证明17β-雌二醇(E_2)在体内有抑制LH分泌的作用。然而E_2对离体垂体前叶分泌LH有无直接抑制作用,尚无一致意见。本文研究了在大鼠动情周期的不同时期,体内内分泌情况不同时,其离体垂体用E_2处理时对促性腺激素分泌的影响。目的在于确定E_2能否直接在垂体水平抑制其基础的和/或受GnRH刺激时的促性腺激素分泌;以及此作用是否受供体动物体内甾体激素变化的影响。实验用成年SD大鼠,每日进行阴道涂片,观察其动情周期的变化。选出处于动情后期(metestrous,M期)及动情前期(proestrous,P期)的大鼠,于上午9∶00断头取出垂体前叶,并收集躯体血,分离血清冰冻,用放射免疫分析方法测定血
17β-estradiol (E_2) has been shown to inhibit LH secretion in vivo. However, there is no consensus on whether E_2 can directly inhibit the secretion of LH in the anterior pituitary gland. In this paper, we studied the effects of E_2 on the secretion of gonadotropin from the isolated pituitary gland in different periods of estrous cycle in rats. The purpose is to determine whether E 2 suppresses its basal and / or gonadotropin secretion directly at the pituitary level when stimulated by GnRH and whether this effect is affected by changes in steroid hormone in the donor animal. Adult Sprague-Dawley rats were used for vaginal smear daily to observe the change of estrous cycle.
