目的 观察口腔鳞癌组织中基质金属蛋白酶 -2 (matrixmetalloproteinase -2 ,MMP -2 )表达及其与微血管密度 (microvesseldensity ,MVD)的关系 ,探讨MMP -2对口腔鳞癌血管生成的作用和意义。方法 应用SABC免疫组织化学法检测 40例口腔鳞癌组织MMP -2表达和MVD ,并分析MMP -2表达与MVD及它们与口腔鳞癌组织临床病理特征之间的关系。结果 癌组织中MMP -2高表达显著高于癌旁组织 ,癌旁组织MMP -2低表达亦明显高于正常组织 (P <0 .0 5 )。MMP -2高表达、MVD与口腔鳞癌病理分级和临床分期无关 ,与口腔鳞癌颈淋巴结转移密切相关 (P <0 .0 5 )。癌组织中MMP -2高表达的MVD均显著高于MMP -2低表达者 (P <0 .0 1)。结论 口腔鳞癌组织MMP -2高表达与MVD及口腔鳞癌颈淋巴结转移有密切关系 ,MMP -2可能是口腔鳞癌的重要促血管生成因子之一
Objective To investigate the expression of matrix metalloproteinase-2 (MMP-2) and its relationship with microvessel density (MVD) in oral squamous cell carcinoma and to investigate the role and significance of MMP-2 in angiogenesis of oral squamous cell carcinoma. Methods SABC immunohistochemistry was used to detect the expression of MMP-2 and MVD in 40 cases of oral squamous cell carcinoma and to analyze the relationship between the expression of MMP-2 and MVD and the clinicopathological characteristics of oral squamous cell carcinoma. Results The high expression of MMP-2 in cancerous tissues was significantly higher than that in paracancerous tissues and the low expression of MMP-2 in paracancerous tissues was also significantly higher than that in normal tissues (P <0.05). The expression of MMP-2 was high. MVD was not associated with pathological grade and clinical stage of oral squamous cell carcinoma, but closely correlated with cervical lymph node metastasis of oral squamous cell carcinoma (P <0.05). The MVD of high expression of MMP-2 in cancer tissue was significantly higher than that of low expression of MMP-2 (P <0.01). Conclusion The high expression of MMP-2 in oral squamous cell carcinoma is closely related to MVD and cervical lymph node metastasis in oral squamous cell carcinoma. MMP-2 may be one of the important angiogenic factors in oral squamous cell carcinoma
