目的研究竹节参不同提取部位预处理对冠脉结扎诱导大鼠急性心肌缺血损伤的保护作用。方法将50只实验大鼠按完全随机设计分组法分为假手术组、模型组和竹节参不同提取部位组;竹节参不同提取部位组大鼠分别灌胃给予竹节参不同提取部位(乙酸乙酯部位、正丁醇部位和水提部位),给药7 d后制作大鼠急性心肌缺血模型,术后12 h记录大鼠心电图,取血,分离血清后测谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性和丙二醛(MDA)、心肌肌钙蛋白I(cTnI)含量,取心脏组织,分别进行心肌组织2,3,5-氯化三苯基四氮唑染色和HE染色。结果竹节参的乙酸乙酯部位组、正丁醇部位组和水提部位均有对抗ST段升高作用,对心肌缺血大鼠心肌梗死面积有较强的降低作用,依次为(10.7±2.4)%、(9.8±2.6)%、(11.0±2.0)%vs(14.8±3.5)%,均具有统计学差异(P<0.05),心肌炎性浸润和溶解坏死现象均有所减轻;在抗氧化损伤和降低cTnI方面,它们能明显增加SOD、GSH-Px酶活性,降低MDA和cTnI含量,与模型组比较,竹节参正丁醇部位均具有统计学差异(P<0.01),水提部位除MDA外,均具有统计学差异(P<0.05,P<0.01)。结论竹节参不同提取部位预处理对冠脉结扎诱导大鼠急性心肌缺血损伤具有较强的保护作用,其中以正丁醇部位最为显著,水提部位次之。
Objective To study the protective effect of different extracts of Scutellaria barbata (pilosula) on acute myocardial ischemia induced by coronary artery ligation in rats. Methods Fifty experimental rats were randomly divided into sham operation group, model group and Scutellaria barbata extract group according to a completely randomized design and grouping method. Rats in different extraction groups were given gavage Ethyl acetate fraction, n-butanol fraction and water extraction site) .After 7 days of administration, acute myocardial ischemia model was made in rats. The electrocardiogram of rats was recorded 12 h after operation, blood was taken and serum was separated and glutathione (GSH-Px), superoxide dismutase (SOD) activity, malondialdehyde (MDA) and cardiac troponin I (cTnI) Triphenyltetrazolium chloride staining and HE staining. Results Ethyl acetate fraction group, n-butanol fraction group and water extract fraction of P.japonicus showed antagonistic effects on ST-segment elevation and myocardial infarction area in myocardial ischemia rats, which were (10.7 ± 2.4%, 9.8 ± 2.6%, 11.0 ± 2.0% vs 14.8 ± 3.5%, respectively. There was a significant difference between the two groups (P <0.05) Oxidative damage and cTnI reduction, they can significantly increase the activity of SOD, GSH-Px and decrease the content of MDA and cTnI. Compared with the model group, the n-butanol fraction of P. japonicus had statistical significance (P <0.01) The sites were statistically different except MDA (P <0.05, P <0.01). Conclusion Pretreatment with different extracts of Scutellaria barbata has a protective effect on acute myocardial ischemia injury induced by coronary ligation in rats. Among them, the n-butanol fraction is the most prominent and the water extract fraction is the second.
