目的:探讨胰岛素样生长因子-1(IGF-1)在大鼠肝纤维化发生中的作用机制。方法:对32只雄性SD大鼠皮下注射40%四氯化碳(CCl4)油剂制备大鼠肝纤维化模型,分别于注射后1,4,8周处理动物,留取肝组织,应用免疫组织化学方法检测大鼠肝组织中IGF-1的表达。结果:免疫组化结果显示,IGF-1主要表达于肝细胞胞质中。注射CCl4诱导后,大鼠肝组织中IGF-1表达较正常对照明显增强(P<0.01,<0.05),且1,4,8周肝组织中IGF-1的表达强度呈明显递增趋势(P<0.01,<0.05)。结论:在大鼠纤维化肝组织中,IGF-1主要表达于肝细胞胞质,可能通过促进肝细胞和肝星状细胞增殖、活化而发挥促纤维化作用。
Objective: To investigate the mechanism of insulin-like growth factor-1 (IGF-1) in the pathogenesis of hepatic fibrosis in rats. Methods: Thirty-two male Sprague-Dawley rats were injected subcutaneously with 40% carbon tetrachloride (CCl4) oil to prepare rat hepatic fibrosis model. Animals were sacrificed at 1, 4 and 8 weeks after injection, Histochemical methods were used to detect the expression of IGF-1 in rat liver tissue. Results: The results of immunohistochemistry showed that IGF-1 was mainly expressed in the cytoplasm of hepatocytes. After injection of CCl4, the expression of IGF-1 in liver tissue of rats increased significantly (P <0.01, <0.05), and the expression of IGF-1 in liver tissues increased significantly <0.01, <0.05). CONCLUSION: IGF-1 is mainly expressed in the cytoplasm of hepatic cells in rat fibrotic liver tissue, and may play a role in promoting fibrosis by promoting the proliferation and activation of hepatocytes and hepatic stellate cells.
