目的:建立HPLC法测定烟酸姜黄素酯原料及其纳米粒中烟酸姜黄素酯的含量并进行方法学考察,为制剂工艺研究提供依据。方法:采用C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.5%醋酸溶液(65∶35)、流速:1.0 ml·min~(-1);检测波长:280 nm;柱温:30℃。结果:烟酸姜黄素酯在5.200~104.000μg·ml~(-1)范围内线性关系良好,r=0.999 4。烟酸姜黄素酯纳米粒及原料平均回收率分别为100.1%,100.9%,RSD分别为1.260%,0.995 0%(n=6)。结论:该分析方法简单,准确,重复性好,可用于烟酸姜黄素酯纳米粒及原料中烟酸姜黄素酯的含量测定及制备工艺研究。
OBJECTIVE: To establish a method for the determination of curcumin in nicotinic acid curcumin and nicotinic acid curcumin ester by HPLC and study the methodology. METHODS: The mobile phase was acetonitrile-0.5% acetic acid (65:35) at a flow rate of 1.0 ml · min -1 using a C 18 column (250 mm × 4.6 mm, 5 μm). The detection wavelength was 280 nm; column temperature: 30 ℃. Results: Curcumin had a good linear relationship in the range of 5.200 ~ 104.000μg · ml ~ (-1), r = 0.999 4. The average recoveries of curcumin niacin nanoparticles and raw materials were 100.1% and 100.9%, respectively, with RSDs of 1.260% and 0.995%, respectively (n = 6). Conclusion: The method is simple, accurate and reproducible. It can be used for the determination of curcumin in nicotinic acid curcumin nanoparticles and its preparation.
