The specific transfer factor(TFHSV-1)obtained from mouse spleensimmunized with herpes simplex virus type Ⅰ(HSV-1)was used in the experimentof ~3H-Leu incorporation of mouse spleen cells.It was observed that TFHSV-1increased obviously ~3H-Leu incorporation of mouse spleen cells in the presence ofHSV-1 antigen.The TFHSV-1 became inactive after incubation at 80℃ for onehour.It was found by UV monitoring and free amino acid analysis that theabsorbance value of inactive TFHSV-1 rose,and the peptides in the heated TFHSV-1degraded partially.This structural change might be closely related to theinactiveness.On the other hand,TFHSV-1 increased non-specifically ~3H-Leuincorporation of mouse spleen cells induced by ConA.These results show that TFHSV-1 has both antigen-specific and non-specificeffects on increasing the protein synthesis of the lymphocytes from mouse spleensin vitro.Some peptides in TFHSV-1 may be the active component responsible for itsantigen-specificity.The ~3H-Leu incorporation method used here is a sensitiveassay for the antigen-specificity of TF in vitro.
