应用c-Ha-ras(6.6Kb)探针检测人类胃癌组织中C-Ha-ras基因序列中5’-CCGG-3’甲基化状态,根据胃组织DNA用HpaII酶切、杂交后所显示的杂交带的位置和出现的频率不同,并且与Mspl酶切、杂交带型比较来判定c-Ha-ras基因甲基化程度的差异。本组胃癌c-Ha-ras基因低甲基化发生率是45%,癌旁组织中发生率是15%,c-Ha-ras基因低甲基化是胃癌发生的重要早期事件。
The c-Ha-ras (6.6Kb) probe was used to detect the 5’-CCGG-3’methylation status of C-Ha-ras gene sequences in human gastric cancer tissues. The DNA was digested with HpaII and digested by gastric tissue. The position and frequency of the hybridization bands shown were different, and the difference in the degree of methylation of the c-Ha-ras gene was determined by comparison with the Mspl cleavage and hybridization bands. The incidence of c-Ha-ras gene hypomethylation in gastric cancer was 45% in this group, and 15% in adjacent tissues. Hypomethylation of c-Ha-ras gene was an important early event in gastric cancer.
