目的探讨丹酚酸B对大鼠非酒精性脂肪性肝炎的疗效。方法雄性SD大鼠36只随机分组,对照组、模型组及丹酚酸B治疗组各12只,对照组给予普通饲料,其余各组均给予高脂饲料。于12周处死模型组。治疗组每天以浓度为1mg/ml的丹酚酸B溶液20ml/kg灌胃,同时继续高脂饲料喂养,对照组以20ml/kg蒸馏水灌胃,同时继续普通饲料喂养。于实验第24周处死剩余实验动物。测定血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、三酰甘油(TG)、总胆固醇(TC)及肝组织丙二醛(MDA)含量,同时观察肝组织病理学改变,免疫组化法检测肝组织TIMP-1的表达,RT-PCR法检测肝组织TGF-β1mRNA的表达。结果与正常对照组相比,模型组肝指数、血清ALT、AST、TG、TC及肝组织MDA水平显著升高(P<0.01),肝组织炎症活动度计分、TIMP-1及TGF-β1mRNA的表达均显著升高(P<0.01)。丹酚酸B治疗组血清ALT、AST、TG、TC及肝组织MDA水平较模型组显著降低(P<0.01),肝组织炎症活动度计分、TIMP-1及TGF-β1mRNA的表达均较模型组显著降低(P<0.01或P<0.05)。结论丹酚酸B对高脂饮食大鼠非酒精性脂肪性肝炎有一定的治疗作用。
Objective To investigate the effect of salvianolic acid B on non-alcoholic steatohepatitis in rats. Methods Thirty - six male Sprague - Dawley rats were randomly divided into control group, model group and salvianolic acid B treatment group. The control group was given normal diet, while the other groups were given high - fat diet. The model group was sacrificed at 12 weeks. The treatment group daily with the concentration of 1mg / ml salvianolic acid B solution 20ml / kg gavage, while continuing to feed high-fat diet, the control group with 20ml / kg distilled water, while continuing to feed ordinary feed. The remaining experimental animals were sacrificed on the 24th week of the experiment. The expression of TIMP-1 in liver tissue was detected by immunohistochemistry. The expression of TGF-β1 mRNA in liver tissue was detected by RT-PCR. Results Compared with the normal control group, the liver index, serum ALT, AST, TG, TC and the level of MDA in the liver tissue of the model group were significantly increased (P <0.01), and the scores of inflammatory activity, TIMP-1 and TGF- Were significantly increased (P <0.01). The levels of ALT, AST, TG, TC and MDA in liver tissue of salvianolic acid B treatment group were significantly lower than those in model group (P <0.01). The expression of TIMP-1 and TGF-β1 mRNA in liver tissues were significantly lower than those in model group Group was significantly lower (P <0.01 or P <0.05). Conclusion Salvianolic acid B has certain therapeutic effect on nonalcoholic steatohepatitis in rats fed with high fat diet.
