[目的]探讨金雀异黄素(Genistein)对胶质瘤细胞株U251MG增殖的影响及可能的作用机制。[方法]用MTT比色法观察不同浓度Genistein在不同作用时间下对U251MG细胞增殖的影响,并通过流式细胞仪和免疫细胞化学染色检测p53(突变型)、c-erbB1、c-myc、cyclinD1蛋白表达。[结果]当Genistein在达到一定浓度或(和)一定时间后,对U251MG细胞增殖均有不同程度的抑制作用,且呈时间剂量依赖性。U251MG细胞经Genistein作用48h后,p53(突变型)、c-myc基因蛋白表达均较对照组明显下降(F=25.766,P﹤0.01;F=22.974,P﹤0.01),且呈剂量依赖性,而c-erbB1、cyclinD1基因蛋白表达无明显变化(F=0.984,P﹥0.05;F=2.416,P﹥0.05)。[结论]Genistein对U251MG细胞的增殖有明显的抑制作用,其抗肿瘤的机制可能与p53(突变型)和c-myc蛋白表达降低有关。
[Objective] To investigate the effect of genistein on the proliferation of glioma cell line U251MG and its possible mechanism. [Method] MTT assay was used to observe the effect of Genistein at different concentrations on the proliferation of U251MG cells. The expressions of p53 (mutant type), c-erbB1, c-myc and c-myc were detected by flow cytometry and immunocytochemistry. cyclinD1 protein expression. [Result] Genistein could inhibit the proliferation of U251MG cells in a dose-dependent and time-dependent manner after reaching a certain concentration or for a certain period of time. The expression of p53 (mutant) and c-myc in U251MG cells treated with Genistein for 48 h decreased significantly (F = 25.766, P <0.01; F = 22.974, P <0.01) There was no significant difference in the expression of c-erbB1 and cyclinD1 (F = 0.984, P> 0.05; F = 2.416, P> 0.05). [Conclusion] Genistein can significantly inhibit the proliferation of U251MG cells, and its anti-tumor mechanism may be related to the decrease of p53 (mutant) and c-myc protein expression.
