目的:研究浙贝母中主要成分生物碱的透皮吸收特性,考察不同浓度溶剂及常用渗透促进剂Azone对其透皮特性的影响。方法:采用药物透皮扩散试验仪进行体外扩散实验,选择离体大鼠背部皮肤为渗透屏障,用UV检测法测定样品中生物碱的含量。结果:浙贝母中主要生物碱的70%乙醇溶液、95%乙醇溶液均可透过大鼠背部皮肤,其差异有统计学意义(P<0.01),但渗透系数较小;Azone对其无显著促渗透作用(P>0.05)。结论:此法将为其经皮给药制剂研究提供可靠实验依据。
OBJECTIVE: To study the transdermal absorption characteristics of alkaloids, the main constituents of Fritillaria cirrhosa, and investigate the effects of different concentrations of solvent and Azone on its transdermal properties. Methods: In vitro diffusion test was carried out by using drug transdermal diffusion tester. The skin of the isolated rat was selected as the permeation barrier. The content of alkaloids in the samples was determined by UV detection. Results: 70% ethanol solution and 95% ethanol solution of the main alkaloids in Fritillaria cirrhosa could permeate the skin of the back of rats, the difference was statistically significant (P <0.01), but the permeability coefficient was small; Significantly promote osmotic effect (P> 0.05). Conclusion: This method will provide a reliable experimental basis for its transdermal drug preparation research.
