目的 观察新二氢吡啶类钙拮抗剂 2 ,6 -二甲基 - 4-(3-硝基苯基 ) - 1 ,4-二氢 - 3,5 -吡啶二羧酸 - 3-甲酯 - 5 -正戊酯(MN92 0 2 )对血栓形成大鼠尾动脉血管平滑肌及内皮功能的影响 .方法 大鼠脚趾 sc注射角叉菜胶制备血栓模型 ,并于注射角叉菜胶前 2 4h,1 h及注射后 2 4h ip MN92 0 2 1μmol·kg- 1 .取尾动脉血栓波及不同区域的三段血管 ,以去甲肾上腺素 (NE) ,乙酰胆碱 (ACh) ,亚硝酸钠 (Na NO2 )为诱导剂 ,进行离体血管条试验 .结果 与正常对照组比较 ,模型组尾动脉对3种诱导剂的反应性显著下降 ,由近端至远端逐渐增加 ,远端血管的舒张反应完全丧失 .与模型组比较 ,MN92 0 2组尾动脉各段对 NE的收缩反应 (mg)有所增强 ,尤以近端最为显著(1 35± 2 5 vs88± 2 0 ,P<0 .0 5 ) ;近端及中段尾动脉血管对ACh诱导的舒张反应显著改善 ,Emax分别为 (34± 3.8) % vs(2 7± 3.1 ) %和 (32± 3.4) % vs (2 4± 4.5 ) % (P<0 .0 5 ) ,对亚硝酸钠诱导的舒张反应有所增加 .结论 MN92 0 2对尾血栓造成的尾动脉血管平滑肌及内皮损伤具有保护作用 ,这有利于其抗血栓形成作用
