目的:探讨脓毒症大鼠肿瘤坏死因子a(TNF a)与肝细胞凋亡的关系以及对肝功能的影响。方法:大鼠随机分为正常对照组和实验组,实验组采用盲肠结扎穿孔术(CLP)致脓毒症大鼠模型,分别于术后30、60、120、180min(CLP术后30、60、120、180 min组)处死动物,测定肝组织和血浆TNF a水平,并检测肝细胞凋亡的情况,同时测定丙氨酸氨基转移酶(ACL)和天冬氨酸氨基转移酶(AST)水平。正常对照组麻醉后不行CLP术,其余同实验组。结果: 与正常对照组相比,CLP术后30、60、120、180 min组肝细胞凋亡数、血清TNF-α、ALT水平均明显升高(P<0.01),有随着时间的延长增加的趋势;与正常对照组相比,CLP术后120、180 min组血清AST含量明显升高(P<0.01);CLP术后大鼠肝组织、TNF-α表达强度随时间延长而有增强的趋势。结论: CLP后大鼠肝脏产生大量TNF-α,与肝细胞凋亡有密切关系,肝功能受到损伤。
Objective: To investigate the relationship between tumor necrosis factor a (TNFa) and hepatocyte apoptosis in septic rats and its effect on liver function. Methods: The rats were randomly divided into normal control group and experimental group. The experimental group was induced by cecal ligation and puncture (CLP) to induce the sepsis model rats at 30, 60, 120 and 180 minutes after operation , 120,180 min group). The levels of TNF-α in liver tissue and plasma were measured and the apoptosis of hepatocytes was detected. The levels of alanine aminotransferase (ACL) and aspartate aminotransferase (AST) Level. Normal control group after anesthesia without CLP surgery, the rest with the experimental group. Results: Compared with the normal control group, the apoptosis of hepatocytes, the levels of serum TNF-α and ALT were significantly increased at 30, 60, 120 and 180 min after CLP (P <0.01) (P <0.01). Compared with the normal control group, serum AST levels in the 120,180 min group increased significantly after CLP (P <0.01); the expression of TNF-α in the liver tissue of the rats after CLP increased significantly with time the trend of. CONCLUSION: A large amount of TNF-α is produced in rat liver after CLP, which is closely related to the apoptosis of liver cells and liver function is impaired.
