目的:探讨逆转录病毒介导的反义bcl2序列对细胞凋亡的诱导作用。方法:PA317细胞包装逆转录病毒,病毒转导Jurkat细胞后用RTPCR及Western印迹法检测bcl2mRNA及蛋白表达水平;用流式细胞仪计数及DNA电泳检测细胞凋亡。结果:转染反义bcl2序列可使内源性BCL2蛋白表达下降,提高白血病细胞株对足叶乙甙(Vp16)的敏感性,促进细胞凋亡。结论:反义bcl2序列能促进Vp16诱导的白血病细胞凋亡,为bcl2反义技术治疗白血病的临床应用提供了实验依据。
Objective: To investigate the effect of retrovirus-mediated antisense bcl2 sequence on apoptosis. METHODS: PA317 cells were packaged with retroviruses, and Jurkat cells were transduced with the virus. The expression of bcl-2 mRNA and protein was detected by RT-PCR and Western blotting. Apoptosis was detected by flow cytometry and DNA electrophoresis. RESULTS: Transfection of antisense bcl2 sequence reduced the expression of endogenous BCL2 protein, increased the sensitivity of leukemic cell lines to Vp16, and promoted apoptosis. Conclusion: Antisense bcl2 sequence can promote the apoptosis of leukemia cells induced by Vp16, and provide experimental basis for the clinical application of bcl2 antisense technology in the treatment of leukemia.
