目的:提取银杏叶聚戊烯醇同系物,对传统工艺中主要的分离、纯化步骤精确并优化。方法:以石油醚为提取溶剂,制得石油醚浸膏中的聚戊烯乙酸酯经水解后转化为聚戊烯醇,然后通过丙酮沉淀及乙醇除杂进一步提高样品纯度,最后过硅胶柱层析得到聚戊烯醇同系物,采用高效液相色谱法(HPLC)测定聚戊烯醇纯度。结果:得到的产物与聚戊烯醇标准品比较,纯度为95.27%。结论:该优化体系提取率高,简单可行,易于工业放大。
Objective: To extract ginkgo polyprenol homologue of Ginkgo biloba, and to separate and purify the major steps in the traditional process accurately and optimally. Methods: The petroleum ether extract was used as solvent to prepare the poly (pentenoyl) acetate in petroleum ether extract which was transformed into polypentenol by hydrolysis. Then the purity of the sample was further increased by acetone precipitation and ethanol impurity removal. Finally, silica gel column Chromatography to obtain polyprenol homologues, using high performance liquid chromatography (HPLC) determination of polyprenol purity. Results: The obtained product showed a purity of 95.27% compared with the standard of polyprenol. Conclusion: The optimized extraction rate is high, simple and feasible, easy to industrial amplification.
