目的:探讨上皮-间质转化(EMT)参与乳腺癌细胞侵袭和转移的过程;双氢青蒿素(DHA)在逆转乳腺癌细胞上皮-间质转化(EMT)过程中的作用,并探讨NF-κB信号分子可能参与逆转EMT过程。方法:利用DHA处理脂多糖(LPS)诱导的乳腺癌细胞株MCF-7,观察细胞形态变化;Western Blot实验及RT-PCR检测DHA干预前后钙黏蛋白(E-cad以及波形蛋白Vimentin)表达水平的变化;Transwell实验检测DHA干预前后乳腺癌细胞侵袭能力的变化;通过Western Blot免疫印迹检测DHA干预前后核转录因子Snail以及P65蛋白的表达变化。结果:DHA可逆转LPS诱导的乳腺癌细胞MCF-7EMT发生,使LPS诱导的EMT相关蛋白E-cad表达上调、Vimentin表达下调;并下调Snail蛋白及NF-κB P65的表达。DHA可抑制发生EMT的乳腺癌细胞运动及侵袭。结论:1LPS可诱导乳腺癌细胞系发生EMT,并增强细胞侵袭能力;2DHA逆转乳腺癌细胞EMT,抑制NF-κB信号通路进而下调核转录因子Snail的表达。
OBJECTIVE: To investigate the involvement of epithelial-mesenchymal transition (EMT) in the invasion and metastasis of breast cancer cells; the role of dihydroartemisinin (DHA) in reversing the epithelial-mesenchymal transition (EMT) in breast cancer cells and to explore the role of NF The -κB signaling molecule may be involved in reversing the EMT process. Methods: DHA was used to treat LPS-induced breast cancer cell line MCF-7 and the morphological changes of cells were observed. The expression of E-cad and vimentin was detected by Western Blot and RT-PCR. The changes of invasion ability of breast cancer cells before and after DHA intervention were detected by Transwell assay. The expressions of Snail and P65 protein were detected by Western Blot before and after DHA intervention. RESULTS: DHA reversed the LPS-induced MCF-7 EMT in breast cancer cells, up-regulated the expression of EMT-related protein E-cadherin and down-regulated the expression of Vimentin, and down-regulated the expression of Snail protein and NF-κB P65 in LPS-induced breast cancer cells. DHA can inhibit the EMT in breast cancer cell motility and invasion. CONCLUSION: 1LPS can induce EMT in breast cancer cell lines and enhance cell invasion ability. 2DHA reverses EMT in breast cancer cells, inhibits NF-κB signaling pathway and down-regulates the expression of nuclear transcription factor Snail.
