目的探索白色念珠菌对罗丹明6G的吸收和泵出的测定意义,并对测定方法进行总结。方法借助2株白色念珠菌敏感株和2株白色念珠菌耐药株,采用多功能微板检测仪、流式细胞仪、荧光显微镜三种仪器,通过定量、半定量和定性的方法测定不同敏感性白色念珠菌对罗丹明6G的吸收和泵出,对定量与半定量结果进行T检验,并与定性结果进行比较。结果不同敏感性白色念珠菌对罗丹明6G的吸收基本一致,实验用耐药菌对罗丹明6G的泵出比敏感菌多;以多功能微板检测仪测定结果为标准,流式细胞仪测定结果与之的一致性为83.3%,荧光显微镜观察结果与之的一致性为91.7%,三种方法具有一致性。结论白色念珠菌对罗丹明6G的吸收和泵出的测定可以为研究新型抗真菌成分的作用机制和真菌耐药逆转剂的作用机制提供工具菌株和可能的研究方向,应根据实验目的和条件选择适当的测定方法。
Objective To explore the determination of Candida albicans absorbance and pump-out of rhodamine 6G, and to summarize the determination methods. Methods Two sensitive strains of Candida albicans and two strains of Candida albicans were used in this study. The multi-functional microplate detector, flow cytometer and fluorescence microscope were used to detect the different sensitive Candida albicans on rhodamine 6G absorption and pumping, quantitative and semi-quantitative results of T test and qualitative results were compared. Results The sensitivity of Candida albicans to rhodamine 6G absorption was basically the same. The resistance of experimental drug-resistant bacteria to rhodamine 6G was more than that of sensitive bacteria. The results of multi-functional microplate detector were the standard. Flow cytometry The results were consistent with the 83.3%, the results of fluorescence microscopy and its consistency was 91.7%, the three methods are consistent. Conclusion The determination of Rhodamine 6G uptake and pump-out by Candida albicans can provide tool strains and possible research directions for studying the action mechanism of novel antifungal components and the mechanism of action of fungal drug resistance reversal agents, and should be selected according to experimental purposes and conditions Appropriate measurement methods.
