目的分析外源性三氯生对胎鼠脑组织抗氧化水平及凋亡蛋白表达影响,并检测母体本身及胎盘的相关指标,探讨其对脑组织发育影响的可能机制。方法将32只妊娠Wistar大鼠随机分为4组:溶剂对照(花生油)组,30、100和300mg/kg剂量组。妊娠6~20天连续灌胃给予相应受试物,孕20天处死,检测相关指标。结果各剂量组胎鼠脑组织总SOD、Cu-Zn SOD、MDA、GSH-Px水平及胶质细胞中Bax、Bcl-2蛋白表达和对照组相比差异有显著性(P<0.05),并且与三氯生的剂量呈一定的相关关系(P<0.05),孕鼠的胸腺系数及TT3水平随三氯生剂量的增加逐渐减小(r=-0.400,P=0.019;r=-0.708,P=0.015),其他指标在各组之间差异不明显(P>0.05)。结论孕鼠妊娠期间接触三氯生对胎鼠脑组织有一定的影响,随着剂量的增大对机体的危害越明显。
OBJECTIVE: To analyze the effect of exogenous triclosan on the antioxidant status and the expression of apoptotic protein in brain tissue of fetal rat and to detect the related indicators of maternal itself and placenta and to explore its possible mechanism of brain tissue development. Methods 32 pregnant Wistar rats were randomly divided into 4 groups: solvent control (peanut oil) group, 30,100 and 300mg / kg dose group. Pregnancy 6 to 20 days continuous gavage given the corresponding test substance, 20 days pregnant were sacrificed to detect the relevant indicators. Results The levels of total SOD, Cu-Zn SOD, MDA, GSH-Px and the expression of Bax and Bcl-2 in glial cells were significantly different among the three groups (P <0.05) And the dose of triclosan (P <0.05). The thymus coefficient and the level of TT3 in pregnant rats decreased with the increase of dosage of triclosan (r = -0.400, P = 0.019; r = -0.708, P = 0.015). There was no significant difference between other groups (P> 0.05). Conclusion Pregnant mice exposed to triclosan during pregnancy have a certain impact on the fetal rat brain tissue, with the dose increased the more obvious harm to the body.
