作者用蔗糖梯度离心分离肝细胞膜,以聚乙二醇为分离剂,建立了大鼠肝细胞膜胰高糖素受体放射分析法。本法的非特异性结合率<5%,特异性结合率25%~35%,批内双管变异为6.83%(n=18),批间重复性符合要求。用受体稀释法和竞争抑制法测定正常及糖尿病大鼠肝细胞膜胰高糖素受体的亲和力均无差异,但糖尿病大鼠肝细胞膜胰高糖素受体浓度较正常大鼠显著增加(P<0.05),尤以低亲和力受体增加明显。作者对受体浓度增加的机理进行了讨论。
The authors used sucrose gradient centrifugation of liver cell membrane, polyethylene glycol as a separating agent, the establishment of the rat hepatocyte membrane glucagon receptor radioactivity assay. The non-specific binding rate of this method is less than 5% and the specific binding rate is 25% -35%. The intra-assay double-tube variation is 6.83% (n = 18). Inter-assay repeatability meets the requirements. There was no difference in the affinity of the glucagon receptor between normal and diabetic rat hepatocytes by both receptor dilution and competition inhibition methods, but the concentration of glucagon receptor in hepatocyte membrane of diabetic rats increased significantly (P <0.05), especially low affinity receptor increased significantly. The authors discuss the mechanism of increased receptor concentration.
