目的观察双黄连粉针剂对多重耐药大肠埃希菌菌体内药物蓄积水平及其细菌结构的影响。方法采用荧光分光光度法观察双黄连粉针剂作用后柔红霉素在多重耐药大肠埃希菌体内聚集水平的改变;透射电镜观察双黄连粉针剂作用后多重耐药大肠埃希菌细菌结构的改变。结果亚抑菌浓度双黄连分别预处理4,6,8 h后,大肠埃希菌菌体内柔红霉素蓄积程度较空白对照增强(P<0.05),其中6~8 h时较为明显;1/2,1/4,1/8最低抑菌浓度(MIC)的双黄连预处理6h后,大肠埃希茵细菌内柔红霉素蓄积程度较空白对照增强(P<0.05),且不同浓度双黄连预处理细菌后柔红霉素蓄积程度改变具有差异(P<0.05)。透射电镜观察,双黄连作用后,多重耐药大肠埃希菌出现分裂相减少、胞膜缺损、胞质流失等改变。结论双黄连粉针剂可以增加柔红霉素在细菌内聚集,呈现量效和时效关系,并且能够破坏大肠埃希菌细胞膜结构的完整性。
Objective To observe the effect of Shuanghuanglian injection on drug accumulation in multidrug-resistant Escherichia coli and its bacterial structure. Methods Fluorescence spectrophotometry was used to observe the changes of daunorubicin accumulation in multidrug-resistant Escherichia coli after Shuanghuanglian injection. The bacterial structure of multidrug-resistant Escherichia coli was observed by transmission electron microscopy change. Results After sub-inhibitory concentration of Shuanghuanglian was pretreated for 4, 6 and 8 h respectively, the accumulation of daunorubicin in Escherichia coli was stronger than that of the blank control (P <0.05), especially in 6 ~ 8 h / 2,1 / 4,1 / 8 minimum inhibitory concentration (MIC) Shuanghuanglian pretreatment 6h, the accumulation of daunorubicin in Escherichia coli than the blank control increased (P <0.05), and different concentrations Shuanghuanglian pretreatment of bacteria daunorubicin accumulation changes have differences (P <0.05). Transmission electron microscopy, Shuanghuanglian effect, multi-drug resistant Escherichia coli decreased mitotic phase, membrane defects, loss of cytoplasm and other changes. Conclusion Shuanghuanglian injection can increase the daunorubicin accumulation in bacteria, showing a dose-effect and time-dependent relationship, and can destroy the integrity of the cell membrane structure of Escherichia coli.
