取CD-1小鼠分别接种曼氏血吸虫尾蚴15~20及80~100条,依次分成轻及重感染两组。由感染9周的小鼠门脉取得成虫,经冷丙酮固定,制成8μm厚的冰冻切片。实验时以针对尾蚴糖蛋白、尾蚴蛋白和雄虫蛋白的单克隆抗体(McAb)以及对照的P3-X63-Ag8腹水液,经硫酸铵沉淀和蛋白A琼脂糖凝胶亲和层析等加以部分纯化,再用四甲基玫瑰红异硫氢酸盐(RIFC)按蛋白:荧光素为2:1的比例结合。抗原抗体均以直接荧光进行检测。肝切片经冷丙酮固定后,PBS洗涤,加用含2mg/ml牛血清白蛋白(BSA)的
