目的通过C57BL/6小鼠干眼模型,探讨中药复方逍生散对干眼杯状细胞的影响及治疗干眼的机制。方法将180只健康C57BL/6小鼠随机分6组:分别为正常组,模型组,生理盐水组,逍生散低剂量、中剂量、高剂量组,每组30只。每天于小鼠后肢根部,皮下注射氢溴酸东莨菪碱造成干眼模型。造模成功后于造模前、造模后、治疗后第4周,测定其泪膜破裂时间、泪液分泌量、角膜染色、结膜杯状细胞数量等情况。结果东莨菪碱皮下肌肉注射造成小鼠干眼模型成功,造模后模型组、生理盐水组、逍生散低剂量组、逍生散中剂量组、逍生散高剂量组在泪液分泌、泪膜破裂时间、杯状细胞数量方面均有明显下降,角膜荧光染色具有明显增加,与造模前有差异(P<0.05),正常组与造模前无明显改变,无统计学意义(P>0.05)。治疗后逍生散中剂量组、逍生散高剂量组在泪液分泌、泪膜破裂时间、角膜荧光染色、杯状细胞数量方面均有明显改善,具有统计学意义(P<0.05)。逍生散低剂量组在泪膜破裂时间有所延长,角膜荧光染色分数有所下降,具有统计学意义(P<0.05)。正常组、模型组、生理盐水组在治疗后有所改变,不具有统计学意义(P>0.05)。结论东莨菪碱皮下肌肉注射可造成小鼠干眼模型。逍生散可增加小鼠泪液分泌量,延长泪膜破裂时间,提高泪膜的稳定性,减少杯状细胞的凋亡,减少眼表损伤等。
Objective To investigate the effect of Xiao-Sheng-San of dry-eye on goblet cells in C57BL / 6 mice and the mechanism of treating dry eye. Methods 180 healthy C57BL / 6 mice were randomly divided into 6 groups: normal group, model group, normal saline group, Xiaozhisan low dose, middle dose and high dose group, 30 rats in each group. Daily mouse hind limbs, subcutaneous injection of scopolamine hydrobromide caused dry eye model. After the success of modeling in the modeling before modeling, 4 weeks after treatment, the determination of tear film rupture time, tear secretion, corneal staining, conjunctival goblet cells and so on. Results Scopolamine intramuscular injection in mice caused by dry eye model was successful. After modeling, model group, saline group, low dosage of Xiao liansan group, Xiao liansan medium dose group and Xiao liansan high dose group had no effect on tear secretion, tear film rupture (P <0.05). There was no significant difference between the normal group and the model group before modeling (P> 0.05), but there was no significant difference between the normal group and the model group (P> 0.05) . After treatment, Xiao-Za-San medium-dose group and Xiao-Sheng-San high-dose group had marked improvement in lacrimal fluid secretion, tear film rupture time, corneal fluorescence staining and goblet cell number, with statistical significance (P <0.05). Xiaozao powder low dose group in tear film time has been extended, corneal fluorescence staining scores decreased, with statistical significance (P <0.05). The normal group, the model group and the saline group changed after the treatment, with no statistical significance (P> 0.05). Conclusion Scopolamine intramuscular injection can cause mouse dry eye model. Xiao-Sheng-Xiao mice can increase the amount of tear secretion, tear film to extend the time to improve the stability of the tear film, goblet cells to reduce apoptosis and reduce ocular surface damage.
