目的研究骨髓间充质干细胞(BMSCs)与骨髓瘤细胞株共培养对成纤维细胞激活蛋白(FAP)、增殖诱导配体(APRIL)和B细胞激活因子(BAFF)表达的影响。方法体外分离、培养BMSCs,并鉴定其成骨分化能力。实验分为BMSCs单独培养的阴性对照组(A组)、BMSCs与U266细胞共培养组(B组)和BMSCs与RPMI8226细胞共培养组(C组)。培养7d和12d后,采用RT-PCR方法检测BMSCs中FAP、APRIL和BAFF mRNA表达水平的变化。结果三组BMSCs形态相似,均具有成骨分化能力。与A组相比,B、C组培养7d后,APRIL、FAP mRNA表达降低(P<0.05),而7d和12d后的BAFF mRNA表达无统计学差异(P>0.05)。结论 BMSCs与骨髓瘤细胞株U266或RPMI8226共培养后,改变FAP和APRIL表达,从而参与骨髓瘤的病理过程。
Objective To investigate the effects of co-culture of bone marrow mesenchymal stem cells (BMSCs) and myeloma cells on the expression of fibroblast activating protein (FAP), proliferation-inducing ligand (APRIL) and B-cell activating factor (BAFF) Methods BMSCs were isolated and cultured in vitro and their osteogenic differentiation ability was identified. The experiment was divided into BMSCs culture alone negative control group (A group), BMSCs and U266 cell co-culture group (B group) and BMSCs and RPMI8226 cell co-culture group (C group). After cultured for 7 days and 12 days, the expression of FAP, APRIL and BAFF mRNA in BMSCs were detected by RT-PCR. Results The three groups of BMSCs were similar in morphology and osteogenic differentiation. Compared with group A, the expression of APRIL and FAP mRNA decreased in group B and group C after 7 days (P <0.05), while there was no significant difference in the expression of BAFF mRNA between group 7 and 12 (P> 0.05). Conclusions After co-cultured with myeloma cell line U266 or RPMI8226, BMSCs can change the expression of FAP and APRIL and participate in the pathological process of myeloma.
