目的:通过运用益气活血法的代表方剂补阳还五汤对动脉粥样硬化模型的干预,观察主动脉组织Rho激酶、VCAM-1及ICAM-1 mRNA的表达、血脂等指标的变化,以揭示该方药抗动脉粥样硬化作用的新靶点和新途径。方法:维生素D3加高脂饮食诱导大鼠动脉粥样硬化模型。60只大鼠用SPSS 13.0软件随机分为6组:正常组、模型组、补阳还五汤低剂量治疗组、补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组。造模成功后干预28 d,检测主动脉Rho激酶、VCAM-1及ICAM-1mRNA表达及血脂水平。结果:高脂饮食加维生素D3可诱导大鼠形成动脉粥样硬化模型,模型组有动脉粥样硬化斑块形成,补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组动脉粥样硬化病变程度明显减轻。Rho激酶、VCAM-1及ICAM-1mRNA表达量水平及血脂水平:模型组与正常组相比,有显著的统计学差异(P<0.01)。补阳还五汤低剂量治疗组与模型组相比有统计学意义(P<0.05),补阳还五汤高剂量治疗组、辛伐他汀对照组、补阳还五汤预防组与模型组相比均有显著的统计学意义(P<0.01),各干预组与正常组相比,有统计学差异(P<0.05)。补阳还五汤预防组、补阳还五汤高剂量治疗组、辛伐他汀对照组各组间无统计学意义(P>0.05)。结论:补阳还五汤可以下调Rho激酶、VCAM-1及ICAM-1mRNA的表达水平、降低血脂,具有抗动脉粥样硬化作用,降低Rho激酶、VCAM-1及ICAM-1mRNA水平可能是其作用机制之一。

Methods: Atherosclerosis model was induced by vitamin D3 plus high fat diet in rats. Sixty rats were randomly divided into six groups with SPSS 13.0 software: normal group, model group, BYHWD low dose treatment group, BYHWD high dose treatment group, simvastatin control group, Soup prevention group. Buyang Huanwu Decoction low-dose treatment group compared with the model group was statistically significant (P <0.05), Buyang Huanwu Decoction high-dose treatment group, simvastatin control group, Buyang Huanwu Decoction and model group (P <0.01). Compared with the normal group, there was a significant difference (P <0.05). Buyang Huanwu Decoction prevention group, Buyang Huanwu Decoction high-dose treatment group, simvastatin control group there was no significant difference between each group (P> 0.05). Conclusion: Buyang Huanwu Decoction can downregulate the expression of Rho kinase, VCAM-1 and ICAM-1mRNA, reduce the level of blood lipids, have anti-atherosclerosis effects and reduce the levels of Rho kinase, VCAM-1 and ICAM-1 mRNA One of the mechanisms.
