目的:观察长毛风毛菊醇提物对α-异硫氰酸萘酯(ANIT)所致肝损伤小鼠的保护作用。方法:将60只小鼠随机分为6组,分别为正常组、联苯双酯组、模型组、长毛风毛菊醇提物低、中、高剂量组(1.0,2.0和4.0 g·kg~(-1))。采用灌胃ANIT花生油溶液的方式复制肝损伤模型,造模48 h后小鼠眼静脉丛取血,测其血清中的丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、胆汁酸(TBA)、总胆红素(TBIL)、肿瘤坏死因子α(TNF-α)的含量;剖取3份肝脏分别用于检测肝组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的含量;分析对TNF-αmRNA表达的影响;观察肝组织病理形态学变化。结果:与模型组相比,长毛风毛菊醇提物中、高剂量组可显著降低肝损伤小鼠血清中ALT,AST,TBA,TBIL和TNF-α的含量(P<0.05,P<0.01);低剂量组只可显著降低肝损伤小鼠血清中TBIL,TNF-α的含量(P<0.05)。长毛风毛菊醇提物各剂量组均可使肝损伤小鼠肝匀浆中SOD水平显著升高,MDA水平显著下降(P<0.05,P<0.01);低、中剂量组能显著影响GSH-Px水平(P<0.01)。各给药剂量组均能影响肝损伤小鼠TNF-αmRNA的表达。病理切片结果显示长毛风毛菊醇提物各剂量组对肝组织的病理变化均有不同程度的改善作用。结论:长毛风毛菊醇提物对ANIT所致小鼠肝损伤有一定的保护作用。
OBJECTIVE: To observe the protective effect of alcohol extract of Humulus vulgaris on hepatic injury induced by α-isothiocyanate (ANIT) in mice. Methods: Sixty mice were randomly divided into 6 groups: normal group, bifendate group, model group, low, medium and high doses of S. furcini extract (1.0, 2.0 and 4.0 g · kg -1). The model of liver injury was reproduced by intragastric administration of ANIT peanut oil solution. After 48 h, the ophthalmic cortex of mice was taken for blood and the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) (TBA), total bilirubin (TBIL) and tumor necrosis factor-α (TNF-α) were detected. Liver tissues were taken for detection of superoxide dismutase (SOD) Malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) were detected. The effect of TNF-αmRNA expression was analyzed. The pathological changes of liver tissue were observed. Results: Compared with the model group, the contents of ALT, AST, TBA, TBIL and TNF-α in serum of liver injury mice significantly decreased (P <0.05, P < 0.01). The low dose group could only reduce the level of serum TBIL and TNF-α in liver injury mice (P <0.05). The content of SOD in liver homogenate of mice with liver injury increased significantly (P <0.05, P <0.01), but low and middle dose groups could significantly affect GSH-Px levels (P <0.01). Each dose group can affect the expression of TNF-αmRNA in liver injury mice. The results of pathology showed that each dose group of S. furcifera alcohol extract had different degrees of improvement on the pathological changes of liver tissue. Conclusion: The extract from Humulus vulgaris has a protective effect on hepatic injury induced by ANIT in mice.
