目的建立Caco-2细胞单层模型并探索其在筛选抗糖尿病药物方面的作用,研究五倍子水提物的降糖作用及机制。方法采用酶抑制剂模型研究五倍子水提物的α-糖苷酶抑制作用;用Caco-2细胞模型进一步研究五倍子水提物对肠上皮细胞麦芽糖酶和蔗糖酶活力的影响以及对葡萄糖转运吸收的影响。结果五倍子水提物能抑制酶抑制剂模型和小肠上皮细胞上的α-糖苷酶;对葡萄糖在Caco-2细胞上的吸收也有一定的抑制作用。结论五倍子抗糖尿病的作用可能主要通过抑制蔗糖酶、麦芽糖酶的活力,作为一种α-葡糖苷酶抑制剂发挥抗糖尿病作用,可能通过抑制葡萄糖吸收而起部分作用。稳定的Caco-2细胞单层模型在筛选抗糖尿病药物方面有重要的意义。
Objective To establish a Caco-2 cell monolayer model and explore its role in the screening of anti-diabetic drugs to study the hypoglycemic effect and mechanism of gallnut water extract. Methods Inhibition of α-glycosidase in aqueous extract of Galla Chinensis was studied by enzyme inhibitor model. The effect of aqueous extract of Galla Chinensis on the activities of maltase and sucrase in intestinal epithelial cells and the effect on the absorption and transportation of glucose were further studied by Caco-2 cell model . Results gallnut water extract can inhibit the enzyme inhibitor model and intestinal epithelial cells α-glycosidase; glucose absorption in Caco-2 cells also have some inhibitory effect. Conclusion The anti-diabetic effect of Chinese nutgall may play an anti-diabetic role as an α-glucosidase inhibitor by inhibiting the activity of sucrase and maltase, and may play a part role by inhibiting glucose absorption. The stable Caco-2 cell monolayer model is of great importance in screening antidiabetic drugs.
