本文以热带假丝酵母作为出发菌株,进行了紫外诱变和驯化,利用木糖生产燃料酒精,以获得高产菌株。通过实验得出:紫外诱变时最佳的菌液稀释倍数为10-4,最佳紫外照射时间为35s,通过三轮的紫外诱变育种进行筛选,获得酒精产量较高的菌株,其中突变株YⅢ27,酒精产率达到了6.78%,木糖利用率为71.1%。对突变株Ⅲ27进行了驯化,驯化后的菌株,酒精产率提高到9.6%,木糖利用率达到了90.3%。
In this paper, Candida tropicalis as a starting strain, UV mutagenesis and domestication, the use of xylose fuel ethanol production to obtain high-yield strains. The results showed that the best bacterium liquid dilution ratio was 10-4 and the best UV irradiation time was 35s when UV mutagenesis. The mutants were screened by three rounds of UV mutagenesis breeding. Strain YIII27, ethanol yield reached 6.78%, xylose utilization rate was 71.1%. The strain Ⅲ27 was domesticated and domesticated. The yield of alcohol increased to 9.6% and the utilization rate of xylose reached 90.3%.
