目的:探讨并观察丹参多糖(SMPS)对体外肝窦内皮细胞(LSEC)窗口的影响。方法:SMPS灌胃大鼠提取的血清干预脂多糖(LPS)刺激的LSEC,检测细胞上清液中的血管活性物质内皮素-1(ET-1)、前列腺素E2(PGE2)的含量,细胞小窝蛋白-1(Cav-1)、α平滑肌肌动蛋白(αSMA)的表达及LSEC窗口数目面积的变化。结果:SMPS能降低细胞上清中ET-1、PGE2的含量,减少Cav-1、αSMA表达,并使肝窦内皮细胞窗孔数目增多、孔径增大。结论:SMPS通过对体外LSEC的干预,改善肝脏循环起到保护肝脏的作用。
Objective: To investigate and observe the effect of Salvia miltiorrhiza polysaccharide (SMPS) on the hepatic sinusoidal endothelial cells (LSEC) window in vitro. Methods: The serum extracted from SMPS was interfered with lipopolysaccharide (LPS) -stimulated LSEC to detect the content of endothelin-1 (ET-1) and prostaglandin E2 (PGE2) in the cell supernatant. Cav-1, α-SMA and the number of LSEC windows were measured. Results: SMPS could decrease ET-1 and PGE2 contents, decrease Cav-1 and αSMA expression, and increase the number of pores and increase the pore size of sinusoidal endothelial cells. Conclusion: SMPS can improve hepatic circulation and protect the liver through the intervention of LSEC in vitro.
