目的:检测原发性舍格伦综合征(primary Sjgren’s syndrome,pSS)患者血浆和血细胞中IFN-α的表达水平,探讨IFN-α在原发性SS发病中的作用。方法:采用ELISA法和实时定量PCR,对37例原发性SS患者和24例非SS患者进行血浆和血细胞中IFN-α水平检测,比较分析其在两组之间的差异。采用SAS6.12软件包进行t检验和Fisher确切概率检验。结果:SS组中,16例(43.24%)患者外周血浆中IFN-α表达阳性,其余21例(56.76%)为阴性;对照组24例患者外周血浆均为阴性表达,2组有显著差异(P=7.02×10-5)。实时定量PCR法检测发现,SS组的IFN-αmRNA表达量显著高于对照组,2组之间有显著差异(P=0.0124)。结论:IFN-α在原发性舍格伦综合征患者中表达升高。
Objective: To detect the expression of IFN-α in plasma and blood cells of patients with primary Sjögren’s syndrome (pSS) and to explore the role of IFN-α in the pathogenesis of primary SS. Methods: Thirty-seven patients with primary SS and 24 patients with non-SS were tested for the level of IFN-α in plasma and blood cells by ELISA and real-time quantitative PCR. The difference between the two groups was analyzed. The SAS6.12 package was used for t test and Fisher exact test. Results: In the SS group, 16 cases (43.24%) had positive peripheral blood plasma IFN-α, and the remaining 21 cases (56.76%) were negative; in the control group, 24 cases were negative for peripheral blood plasma, P = 7.02 × 10-5). Real-time quantitative PCR assay showed that the expression of IFN-αmRNA in SS group was significantly higher than that in control group, there was a significant difference between the two groups (P = 0.0124). Conclusion: IFN-α is elevated in patients with primary Sjogren’s syndrome.
