目的探讨丝氨酸/苏氨酸激酶15(Aurora-A)在星形细胞瘤中的表达以及与临床病理指标的关系。方法用免疫组化方法(两步法)检测33例星形细胞瘤和10例非肿瘤脑组织石蜡切片中Aurora-A蛋白的表达情况,用Western Blot进一步检测Aurora-A蛋白在20例星形细胞瘤及20例非肿瘤脑组织新鲜标本中的表达,分析其与星形细胞瘤临床病理指标的关系。结果Aurora-A蛋白在33例石蜡切片中的表达阳性率为60.6%,10例非肿瘤脑组织石蜡切片中均不表达,两者差异有显著性(P<0.01);在20例星形细胞瘤新鲜标本中的表达阳性率为100%,而在所有非肿瘤脑组织新鲜标本中均不表达,两者差异有显著性(P<0.01)。利用t检验分析可知不同级别星形细胞瘤Aurora-A蛋白的表达差异具有显著性(P<0.05),不同年龄、性别之间表达差异无显著性。结论Aurora-A蛋白表达强度可作为判定星形细胞瘤恶性程度新的指标。
Objective To investigate the expression of Aurora-A in astrocytomas and its relationship with clinicopathological parameters. Methods The expression of Aurora-A protein in 33 cases of astrocytoma and 10 cases of non-tumor brain tissue paraffin sections was detected by immunohistochemical method (two-step method). Aurora-A protein was further detected by Western Blot in 20 cases Cell tumor and 20 cases of non-tumor brain tissue fresh specimens of the expression of astrocytoma and its clinical and pathological indicators. Results The positive rate of Aurora-A protein in paraffin-embedded sections of 33 cases was 60.6%, while it was not found in 10 cases of non-tumor paraffin sections (P <0.01). In 20 cases of astrocytes The positive rate of fresh tumor specimens was 100%, but not in all non-tumor brain tissue fresh specimens, the difference was significant (P <0.01). The results of t-test showed that the expression of Aurora-A in different grades of astrocytomas was significantly different (P <0.05). There was no significant difference between different age and sex. Conclusion Aurora-A protein expression intensity can be used as a new indicator to determine the degree of malignancy of astrocytoma.
