目的:观察产后抑郁症(PPD)模型大鼠Treg和Th17细胞比例的变化,从T细胞免疫入手,探讨参芪解郁方调节PPD Treg/Th17平衡的作用机制。方法:90只SD雌性大鼠随机分为正常组、假手术、模型组、中药组和西药组,每组6只,分设灌胃1、2、4周3个时点。采用流式细胞术检测3个时点各组大鼠外周血Treg细胞和Th17细胞比例,并计算Treg/Th17比值。结果:各时点模型组Treg细胞数量较正常组降低,1周和2周较同时点下降明显(P<0.05,P<0.01);1周、2周模型组Th17细胞数量较正常组明显下降(P<0.01),而4周模型组Th17细胞数量较正常组明显上升(P<0.01),2周西药组较模型组升高明显(P<0.01),4周中药组较模型组降低明显(P<0.01);1周、2周模型组Treg/Th17较正常组明显升高(P<0.01),而4周模型组Treg/Th17较正常组明显降低(P<0.01),4周中药组较模型组升高明显(P<0.01)。结论:Treg/Th17平衡参与PPD发生发展,参芪解郁方通过调控Treg/Th17的失衡状态发挥效用,优于西药。
Objective: To observe the changes of Treg and Th17 cell ratio in postnatal depression (PPD) model rats. To study the mechanism of Shenqi Jieyu prescription regulating the balance of PPD Treg / Th17 cells from T cell immunity. Methods: Ninety SD female rats were randomly divided into normal group, sham operation group, model group, TCM group and western medicine group, with 6 rats in each group. Flow cytometry was used to detect the percentage of Treg cells and Th17 cells in peripheral blood of three groups at each time point and the ratio of Treg / Th17 was calculated. Results: The number of Treg cells in the model group at each time point was lower than that in the normal group (P <0.05, P <0.01), and the number of Th17 cells in the model group at 1 week and 2 weeks was significantly lower than that of the normal group (P <0.01). The number of Th17 cells in 4-week model group was significantly higher than that in normal group (P <0.01), and increased significantly in Western medicine group compared with model group at 2 weeks (P <0.01) (P <0.01). Treg / Th17 in model group at 1 week and 2 weeks were significantly higher than those in normal group (P <0.01), while Treg / Th17 at 4 weeks was significantly lower than that of normal group (P <0.01) Group than the model group increased significantly (P <0.01). Conclusion: The balance of Treg / Th17 is involved in the development of PPD. Shenqi Jieyu decoction exerts its effect by regulating the imbalance of Treg / Th17 and is superior to western medicine.
