目的:探讨血府逐瘀汤及其拆方对动脉粥样硬化血管平滑肌细胞丝裂原活化蛋白激酶(MAPK)负反馈因子丝裂原蛋白激酶磷酸酶(MKP-1 mRNA)表达的影响。方法:采用高脂饲料复制动脉粥样硬化模型,原位杂交法检测主动脉MKP-1mRNA的表达。结果:模型组与空白组比较,模型组MKP-1 mRNA表达显著增高,差异有统计学意义(P<0.01);各药物组与模型组比较,血府组和四逆组MKP-1mRNA表达增多(分别为P<0.01,P<0.05),差异有统计学意义;桃红组差异无统计学意义(P>0.05)。结论:血府逐瘀汤可以有效升高血管平滑肌MKP-1mRNA的表达,可能是其抗动脉粥样硬化血管平滑肌细胞增殖的机理之一。
Objective: To investigate the effect of Xuefu Zhuyu Decoction and its disassembled prescriptions on the expression of mitogen-activated protein kinase (MAPK) negative feedback factor (MAPK) phosphatase (MKP-1 mRNA) in atherosclerotic vascular smooth muscle cells. Methods: Atherosclerosis model was reproduced by high fat diet and the expression of MKP-1 mRNA in aorta was detected by in situ hybridization. Results: The expression of MKP-1 mRNA in the model group was significantly higher than that in the control group (P<0.01). The expression of MKP-1 mRNA was increased in the Xuefu and Sini groups compared with the model group. (P<0.01, P<0.05, respectively), the difference was statistically significant; there was no statistically significant difference in the peach-red group (P>0.05). Conclusion: Xuefu Zhuyu decoction can effectively increase the expression of MKP-1 mRNA in vascular smooth muscle, which may be one of the mechanisms of its anti-atherosclerotic vascular smooth muscle cells proliferation.
