目的:观察无患子皂苷(Sapindoside)对大鼠心肌缺血再灌注模型大鼠心肌酶学及胞浆蛋白NF-κβ表达的影响。方法:通过结扎大鼠左冠状动脉前降支以制备心肌缺血再灌注损伤模型,用比色法检测大鼠心肌匀浆中钠-钾-三磷酸腺苷酶(Na+-K+-ATPase)、钙-三磷酸甘酶(Ca2+-ATPase)和肌酸激酶(CK)的含量;显微镜下观察心肌坏死程度;用酶联免疫吸附法(ELISA)检测核因子κβ(NF-κβ)在心肌细胞中的表达。结果:检验显示,无患子皂苷能提高心肌中Na+-K+-ATPase、Ca2+-ATPase活性,降低心肌中CK的含量;与假手术相比,模型组大鼠心肌缺血再灌注后胞浆蛋白NF-κβ表达明显减少,提示无患子皂苷可升高大鼠心肌缺血再灌注后胞浆蛋白NF-κβ的表达。结论:无患子皂苷对大鼠心肌缺血再灌注损伤有一定的保护作用。其作用机制可能是其能增加缺血区心肌能量物质含量,保护AT-Pase活性,改善能量代谢障碍,抑制NF-κβ的活性,从而对心肌缺血再灌注损伤起到保护作用。
Objective: To observe the effect of Sapindoside on myocardial enzymes and the expression of cytoplasmic protein NF-κβ in rats with myocardial ischemia-reperfusion injury. Methods: The model of myocardial ischemia-reperfusion injury was established by ligating left anterior descending coronary artery in rats. Conclusion: Saponins from Saponins have a protective effect on myocardial ischemia-reperfusion injury in rats. Its mechanism may be that it can increase myocardial ischemic myocardial energy and substance content, protect AT-Pase activity, improve energy metabolism, inhibit the activity of NF-κβ, and thus play a protective effect on myocardial ischemia-reperfusion injury.
