本实验用抗生物素蛋白-生物素过氧化酶复合体方法(S-P)检测了30例转移及非转移鼻咽癌组织中肿瘤细胞的CD44表达,同时检测了将克隆株(CNE-2Z-H5)裸鼠体内连续传代获得的裸鼠移植瘤组织和回复培养细胞CD44粘附分子的表达。30例鼻咽癌标本中有18例表达阳性,CD44表达与肿瘤的分化密切相关,在中等程度到低分化肿瘤组织的肿瘤细胞表达强阳性,肿瘤的侵袭和转移密切相关,其中转移组阳性表达明显高于非转移组(P<0.05)。不同代之间裸鼠移值瘤和回复培养的肿瘤细胞中CD44粘附分子的表达有差异。其中第四代裸鼠移植瘤和回复培养的肿瘤细胞CD44粘附分子的表达明显高于原代移植瘤及原代细胞(P<0.05)。裸鼠移植瘤中CD44粘附分子的表达与肿瘤转移率成正相关关系。在鼻咽癌组织中CD44强表达可能与肿瘤低分化和转移有关。CD44粘附分子的表达与鼻咽癌裸鼠移植瘤转移关系密切。
In this experiment, the CD44 expression of tumor cells in 30 metastatic and non-metastatic nasopharyngeal carcinoma tissues was detected using the avidin-biotin peroxidase complex method (S-P). The cloned strain (CNE-2Z) was also tested. -H5) Expression of CD44 adhesion molecules in nude mouse xenograft tumor tissue and recovery cultured cells obtained by serial passage in nude mice. Thirty out of 30 nasopharyngeal carcinoma specimens were positive. The expression of CD44 was closely related to the differentiation of tumors. Strong positive expression was observed in tumor cells from moderate to poorly differentiated tumors. Invasion and metastasis of tumors were closely related, with positive expression in metastatic group. Significantly higher than the non-metastatic group (P<0.05). The expression of CD44 adhesion molecules differed between nude mouse transplanted tumors and regenerated cultured tumor cells between different generations. Among them, the expression of CD44 adhesion molecules in the fourth-generation nude mice transplanted tumors and regenerated cultured tumor cells was significantly higher than that of primary transplanted tumors and primary cells (P<0.05). There was a positive correlation between the expression of CD44 adhesion molecules and tumor metastasis in nude mice. The strong expression of CD44 in nasopharyngeal carcinoma may be related to the poor differentiation and metastasis of the tumor. The expression of CD44 adhesion molecules is closely related to the metastasis of nasopharyngeal carcinoma in nude mice.
