目的:研究苦瓜总皂苷对IgA肾病大鼠转化生长因子-β(Transforming growth factor-β,TGF-β)表达的影响。方法:随机选取48只雄性SD大鼠,所有大鼠均于适应性喂养1周后随机分为空白对照组、IgA肾病组以及苦瓜总皂苷治疗组,n=16,经典造模法造模成功后,治疗组给予苦瓜总皂苷20 mg/(kg·d)治疗,IgA肾病组和空白组给予等量生理盐水灌胃1 m L/(kg·d),分别在治疗4周和8后,每组处死一半大鼠,观察血清尿素氮、血肌酐、24 h蛋白尿和血清中白介素-2(IL-2)的白介素-6(IL-6)的含量,此外HE染色分析其病理学,Western Blot和RT-PCR检测TGF-β_1和RGC-32表达。结果:IgA肾病组和治疗组所有大鼠均造模成功,主要表现为有明显肉眼或镜下血尿;此外与空白组相比,IgA肾病组和治疗组血清中BUN、24 h尿蛋白、SCr、IL-2和IL-6明显升高(P<0.05),苦瓜总皂苷治疗4周后,上述指标均明显降低(P<0.05),治疗8周后进一步降低;病理学检查显示空白组纹理结构清晰,而IgA肾病组有明显增生反应,苦瓜总皂苷对其有一定的改善。Western Blot和RT-PCR结果显示与空白组相比,IgA肾病组肾脏组织中TGF-β_1表达明显降低(P<0.05),治疗8周后进一步降低,但其对RGC-32基本没有改善作用。结论:苦瓜总皂苷可通过降低肾脏组织中TGF-β_1的表达,进而有助于炎性因子和肾功能的改善,防止其疾病的进一步进展。
Objective: To study the effect of Momordica charantia polysaccharides on the expression of transforming growth factor-β (TGF-β) in rats with IgA nephropathy. Methods: Forty-eight male Sprague-Dawley rats were randomly selected. All rats were randomly divided into blank control group, IgA nephropathy group and total saponin-treated group with n = 16 one week after adaptive feeding. The treatment group was treated with 20 mg / (kg · d) bitter gourd total saponin, IgA nephropathy group and blank group were given equal volume of normal saline gavage 1 m L / (kg · d), respectively, after 4 weeks and 8 weeks, Half of rats in each group were sacrificed and serum urea nitrogen, serum creatinine, 24 h proteinuria and interleukin-6 (IL-6) in serum were measured. HE staining was used to analyze the pathology, Western Blot and RT-PCR detection of TGF-β 1 and RGC-32 expression. Results: All the rats in the IgA nephropathy group and the treatment group were successfully established. The main manifestations were obvious macroscopic or microscopic hematuria. Compared with the blank group, serum levels of BUN, 24 h urinary protein, SCr , IL-2 and IL-6 were significantly increased (P <0.05). After 4-week treatment, the above indexes were significantly decreased (P <0.05) Structure clear, and IgA nephropathy group had obvious proliferative response, bitter gourd total saponin to its certain improvement. The results of Western Blot and RT-PCR showed that the expression of TGF-β 1 in IgA nephropathy group was significantly lower than that in blank group (P <0.05), and decreased further after 8 weeks of treatment. However, RGC-32 had no effect on the expression of TGF-β 1. Conclusion: The total saponin of Momordica charantia can help to improve the inflammatory factor and renal function by reducing the expression of TGF-β 1 in the kidney and prevent its further progress.
