选取SD大鼠48只,随机分为6组,即空白对照组,气道神经源性炎症模型组,惠菲宁对照组及蝉芩颗粒低、中、高剂量组,每组8只。HE染色观察肺组织病理变化;ELISA法检测血清中速激肽(SP、NKA、NKB)及降钙素基因相关肽(CGRP)的含量;免疫组化法检测肺泡及支气管中SP、NKA的蛋白表达。观察蝉芩颗粒对大鼠气道神经源性炎症的干预作用。治疗后蝉芩颗粒低剂量组、中剂量组、高剂量组、惠菲宁组与模型组相比,血清中速激肽SP、NKA、NKB值下降(P<0.05),各治疗组间血清中速激肽含量比较,差异无统计学意义(P>0.05)。治疗后蝉芩颗粒低剂量组、中剂量组、高剂量组、惠菲宁组与模型组相比,血清中CGRP值下降(P<0.05),蝉芩颗粒高剂量组血清中CGRP值低于惠菲宁组(P<0.05)。治疗后蝉芩颗粒中、高剂量,惠菲宁组中支气管SP的表达低于模型组(P<0.05),说明中、高剂量的蝉芩颗粒、惠菲宁可抑制大鼠气道神经源性炎症模型支气管中SP的蛋白表达。治疗后蝉芩颗粒中、高剂量、惠菲宁组中肺泡NKA的表达低于模型组(P<0.05),说明蝉芩颗粒中、高剂量、惠菲宁可抑制大鼠气道神经源性炎症模型肺泡中NKA的蛋白表达。治疗后蝉芩颗高剂量组支气管中NKA的表达低于模型组(P<0.05),说明蝉芩颗粒高剂量组可抑制大鼠气道神经源性炎症模型支气管中NKA的蛋白表达。
Forty-eight SD rats were randomly divided into six groups, namely blank control group, model of airway neurogenic inflammation, Huifin Ning control group and low, medium and high dose group of Chanchuang Granules, with 8 rats in each group. The pathological changes of lung tissue were observed by HE staining. The contents of tachykinins (SP, NKA, NKB) and calcitonin gene related peptide (CGRP) in serum were detected by ELISA. The SP and NKA protein in alveolar and bronchial were detected by immunohistochemistry expression. To observe the intervention of Chanchuang Granule on airway neurogenic inflammation in rats. Compared with the model group, the levels of tachykinins SP, NKA and NKB in the low-dose group, the medium-dose group, the high-dose group and the Huifining group in the treated group were significantly lower than those in the model group (P <0.05) There was no significant difference in middle-speed kinin content (P> 0.05). Compared with the model group, CGRP in serum of Ciclofen pellet low dose group, medium dose group, high dose group and Huifenin group were decreased (P <0.05) Hui Feining group (P <0.05). After treatment, the expression of SP in the bronchi in the middle and high doses of Chanqin Granules was higher than that in the untreated group (P <0.05), indicating that medium and high doses of Chanchuang Granules and Huifining can inhibit airway neurogenicity Protein expression of SP in bronchial inflammation model. After treatment, the expression of NKA in alfalfa at the middle and high dose of Qicheng Granules was lower than that in model group (P <0.05), indicating that mid and high dosage of Chanqin Granules could inhibit the airway neurogenic inflammation in rats Expression of NKA protein in model alveoli. After treatment, the expression of NKA in bronchial of high dose QCHQ group was lower than that in model group (P <0.05), indicating that high dose of Chanchuang Granule could inhibit the expression of NKA in bronchus of airway neurogenic inflammation rat model.
